AKTIVITAS PENGHAMBAT ALFA AMILASE (a-AMYLASE INHIBITOR) HIDROLISAT GELATIN TULANG ITIK

Diabetes mellitus (DM) merupakan penyakit metabolik yang ditandai dengan menurunnya produksi insulin. DM ada 2 tipe dan yang sering diderita oleh masyarakat adalaha tipe II yaitu kadar gula darah melebihi batas normal yaitu 70-130 mg/dL. Terapi obat untuk penyembuhan DM memiliki efek negatif sehingga perlu dicari alternatif secara non farmakologi. Beberapa penelitian melaporkan bahwa peptida yang dihasilkan dari hidrolisis protein memiliki aktivitas inhibitor enzim alfa amilase dan alfa glukosidase. Tujuan dari penelitian ini yaitu untuk mengetahui proses hidrolisis gelatin menggunakan enzim sistein protease serta aktivitas penghambatan alfa amilase. Penelitian ini dibagi menjadi 2 tahap yaitu pertama ekstraksi gelatin kedua hidrolisis gelatin serta uji penghambatan alfa amilase. Penelitian menggunakan rancangan acak lengkap pola searah dengan 3 kali ulangan. Parameter penelitian meliputi total protein terlarut, derajat hidrolisis, serta aktivitas penghambatan alfa amilase. Hasil penelitian menunjukan perbedaan perlakuan waktu hidrolisis berpengaruh nyata (P<0.05) terhadap seluruh parameter. Total protein tertinggi terdapat pada waktu hidrolisis 180 menit yaitu sebesar 463,27 mg/ml, sedangkan derajat hidrolisis tertinggi terdapat pada waktu hidrolisis 240 menit yaitu sebesar 20,49%. Nilai IC50 terhadap alfa amilase dari perlakuan waktu hidrolisis 240 menit sebesar 5,92 mg/ml. Penelitian selanjutnya perlu dilakukan optimasi kembali proses hidrolisis.
